Introduction

Research samples Discover CpG loci associated with mortality Correlation Analysis Samples are randomly selected to participate in the study, and relevant personal information is recorded in detail, including gender, age, disease, etc. Sample forms such as blood, saliva, and cells can be selected for methylation measurement. We isolate sample DNA and measure genome-wide DNA methylation levels and perform data pre-processing to summarize the assessment of DNA methylation levels at each CpG locus and identify loci associated with mortality. We use the discovery of relevant CpG loci to construct DNA methylation models that can estimate the links between age acceleration and mortality, as well as association with other factors influencing death.

Data Analysis

  • DNA methylation assessment. We provide processing and analysis of the methylation data, detecting P-values > 0.01 and deletion of data with deletion values > 5%, and performing genome-wide screening for specific CpG loci for the rest.
  • Statistical analysis. We validate genome-wide specificity of CpG, with CpG in the validation group with significance < 0.05 considered as mortality-associated loci, as well as exploring the factors influencing DNA methylation associated with mortality with multiplex testing correction.
